ALFA-SEQ Advanced Soil DNA Kit採用矽膠柱純化技術,適用於從AMD底泥、重金屬汙染土壤、沙漠幹土、底泥等各種土壤樣品中提取高產量高純度的總DNA;結合獨有的兩步去雜的技術,可高效地吸附土壤樣品中的腐殖酸和其它抑制因子。本試劑盒步驟簡單,可以在1小時內完成微量DNA的提取純化,無需液氮研磨,無需加熱步驟,無需酚氯仿等試劑,安全快速可靠。純化的DNA可直接用於PCR、Southern Blot、酶切、一代測序和二代測序等實驗!
該研究設置普通堆肥和高溫堆肥兩種堆肥試驗,使用ALFA-SEQ Advanced Soil DNA Kit 從0.25 g堆肥樣品中提取出高質量的DNA,用於下遊實驗;對比分析發現高溫堆肥可大大減少N2O的排放,研究結果於10月21日發表在中科院一區期刊《Bioresource Technology》上,崔鵬博士為第一作者。

本研究首次報導了高溫堆肥(HTC)比傳統堆肥(TC)可減少90%的N2O累積排放量。HTC中NO2--N和NO3--N的濃度明顯低於TC,這可能是減少N2O排放的主要原因。此外,還發現HTC中N2O排放量的減少主要是由於抑制了細菌amoA 和norB 基因的豐度,從而分別降低了硝化速率和控制N2O的形成。偏最小二乘路徑模型分析表明,高溫是降低HTC N2O排放的關鍵因素,而物理化學性質是提高TC N2O排放的主要因素。以上表明,HTC是一種可降低糞便堆肥中N2O排放的環境友好型技術,前景廣闊。
