近日,清華大學婁志勇、饒子和等研究人員合作揭示SARS-CoV-2複製和轉錄複合體冷凍電鏡結構。該項研究成果於2020年11月14日在線發表在《細胞》雜誌上。
研究人員報導了SARS-CoV-2複製和轉錄複合物(RTC)向帽結構合成過渡的結構快照。研究人員確定了由nsp7-nsp82-nsp12-nsp132-RNA和單個RNA結合蛋白nsp9組裝的擴展RTC原子冷凍電鏡結構。Nsp9與nsp12(RdRp)NiRAN緊密結合,使nsp9 N末端插入nsp12 NiRAN的催化中心,從而抑制活性。
研究人員還顯示,nsp12 NiRAN具有鳥苷酸轉移酶活性,可催化帽核結構(GpppA)的形成。錨定模板RNA 5'延伸的nsp13的方向顯示出顯著的構象變化,導致其ZBD的鋅指3插入配對的模板引物RNA的較小凹槽中。這些結果說明了RTC處於mRNA合成的中間狀態,為理解RTC架構鋪平了道路,並為抗病毒開發提供了目標。
據悉,SARS-CoV-2 mRNA的轉錄需要RTC促進依次的反應。

Title: Cryo-EM structure of an extended SARS-CoV-2 replication and transcription complex reveals an intermediate state in cap synthesis

Abstract: Transcription of SARS-CoV-2 mRNA requires sequential reactions facilitated by the replication and transcription complex (RTC). Here, we present a structural snapshot of SARS-CoV-2 RTC as it transition towards cap structure synthesis. We determine the atomic cryo-EM structure of an extended RTC assembled by nsp7-nsp82-nsp12-nsp132-RNA and a single RNA binding protein nsp9. Nsp9 binds tightly to nsp12 (RdRp) NiRAN, allowing nsp9 N-terminus inserting into the catalytic center of nsp12 NiRAN, which then inhibits activity. We also show that nsp12 NiRAN possesses guanylyltransferase activity, catalyzing the formation of cap core structure (GpppA). The orientation of nsp13 that anchors the 5』 extension of template RNA shows a remarkable conformational shift, resulting in zinc finger 3 of its ZBD inserting into a minor groove of paired template-primer RNA. These results reason an intermediate state of RTC towards mRNA synthesis, pave a way to understand the RTC architecture, and provide a target for antiviral development.
