抑癌蛋白Rb可調控細胞大小

美國史丹福大學Jan M. Skotheim小組發現,細胞生長會稀釋細胞周期抑制蛋白Rb從而觸發細胞分裂。該項研究成果發表在2020年7月24日出版的《科學》雜誌上。
研究人員發現,在細胞分裂周期G1階段中,細胞生長會稀釋細胞周期抑制蛋白視網膜母細胞瘤蛋白(Rb),從而觸發人類細胞分裂。RB過表達增加細胞大小和G1持續時間,而RB缺失會降低細胞大小並消除出生時細胞大小和G1階段持續時間之間的逆相關性。
因此,在G1期通過細胞生長來稀釋Rb是人們長期尋找的細胞大小穩態調控分子機制之一。
據了解,細胞大小是細胞生理學的基礎。例如,細胞大小決定了細胞器和細胞內運輸的空間尺度,從而影響生物合成。儘管人們已經發現了一些影響哺乳動物細胞大小的基因,但是細胞生長驅動細胞分裂的分子機制仍然難以捉摸。

Title: Cell growth dilutes the cell cycle inhibitor Rb to trigger cell division

Abstract: Cell size is fundamental to cell physiology. For example, cell size determines the spatial scale of organelles and intracellular transport and thereby affects biosynthesis. Although some genes that affect mammalian cell size have been identified, the molecular mechanisms through which cell growth drives cell division have remained elusive. We show that cell growth during the G1 phase of the cell division cycle dilutes the cell cycle inhibitor Retinoblastoma protein (Rb) to trigger division in human cells. RB overexpression increased cell size and G1 duration, whereas RB deletion decreased cell size and removed the inverse correlation between cell size at birth and the duration of the G1 phase. Thus, Rb dilution through cell growth in G1 provides one of the long-sought molecular mechanisms that promotes cell size homeostasis.
