上海交大瑞金醫院揭示NKTCL的基因組和轉錄組學表徵

上海交通大學瑞金醫院趙維蒞、陳賽娟以及黃金豔課題組合作取得一項新成果。他們揭示了自然殺傷T細胞淋巴瘤(NKTCL)的基因組和轉錄組學表徵。相關論文於2020年3月16日發表在《癌細胞》雜誌上。
NKTCL是非霍奇金淋巴瘤的侵襲性和異質性實體,與EBV感染密切相關。
為了根據基因組結構改變和愛潑斯坦-巴爾病毒(EBV)序列鑑定NKTCL的分子亞型,研究人員對128例新診斷的NKTCL組織活檢樣本進行了多組學研究,並定義了三種突出的亞型,它們在起源細、EBV基因表達、轉錄特徵,以及對基於天冬醯胺酶的治療方案和靶向治療應答都不相同。
這一新研究確定了EBV相關發病機理的分子網絡,並提出了NKTCL的潛在臨床治療策略。

Abstract: Natural killer/T cell lymphoma (NKTCL) is an aggressive and heterogeneous entity ofnon-Hodgkin lymphoma, strongly associated with Epstein-Barr virus (EBV) infection.To identify molecular subtypes of NKTCL based on genomic structural alterations andEBV sequences, we performed multi-omics study on 128 biopsy samples of newly diagnosedNKTCL and defined three prominent subtypes, which differ significantly in cell oforigin, EBV gene expression, transcriptional signatures, and responses to asparaginase-basedregimens and targeted therapy. Our findings thus identify molecular networks of EBV-associatedpathogenesis and suggest potential clinical strategies on NKTCL.
